Zinc, magnesium and testosterone: what the two ZMA trials actually found
An industry-funded study said a zinc-magnesium blend raised free testosterone. A larger independent one, run eight weeks later, found nothing at all.
Zinc and magnesium are the two minerals that show up in almost every testosterone stack, usually bundled together as ZMA. The bundle exists because of one study, and it is still sold because most people have not read the second one.
The study that launched the product
In 2000, researchers reported that competitive football players taking a nightly combination of zinc monomethionine aspartate, magnesium aspartate and vitamin B6 for eight weeks saw free testosterone rise from about 132 to 176 pg/mL, while a placebo group's free testosterone fell. IGF-I moved the same way and strength measures favoured the supplement group.
Two things about that study matter. It was small — twelve men on the supplement, fifteen on placebo. And the affiliation list is not neutral: one of the authors worked for the company that developed the formulation, and the laboratory named in the paper is the one associated with it. That does not make the result false. It makes it a claim that needed an independent test.
The independent test
Four years later, a Baylor University group ran that test properly. Forty-two resistance-trained men, matched for fat-free mass, randomly assigned in double-blind fashion to ZMA or a placebo, taken 30 to 60 minutes before sleep through eight weeks of standardised resistance training. Body composition by DXA, strength and endurance testing, anaerobic power, and blood analysis at weeks 0, 4 and 8.
The result: zinc levels rose non-significantly, and no significant differences appeared between groups in anabolic or catabolic hormone status, body composition, bench press or leg press strength, muscular endurance, or anaerobic capacity. The authors' conclusion was that ZMA during training does not appear to enhance training adaptations.
Same ingredients, better-powered design, opposite answer. When the two are laid side by side, the larger independent trial carries more weight, and it found nothing.
The honest nuance about deficiency
Neither study settles the underlying minerals, because both were done in people who were probably not deficient.
There is a reasonable body of observational and review work suggesting magnesium status relates to testosterone bioactivity and physical function in men, and that the relationship is more interesting than a simple cause-and-effect. Systematic reviews of mineral supplementation in athletes tend to land in the same place: benefits show up where there was a deficit to correct, and not much else.
That distinction is the whole story. If your diet is low in these minerals — heavy on processed food, low in meat, shellfish, legumes, nuts, seeds and leafy greens, or you are a restrictive eater — correcting that plausibly helps, and would help your health regardless of what it does to hormones. If you already eat well, adding more has no evidence of benefit and its own costs.
Where that leaves you
- Food first, and there is no contest about it. Zinc is concentrated in oysters, red meat, pumpkin seeds and lentils; magnesium in leafy greens, nuts, seeds and whole grains. A diet that supplies these reliably is doing the job a pill is supposed to imitate.
- Test before you supplement. If you suspect a deficiency, a blood test settles it far more cheaply than a year of guessing.
- More is not better with zinc. Habitual high zinc intake interferes with copper absorption and can cause genuine harm. There are established upper limits for a reason.
- Magnesium is usually a sleep story, not a testosterone story. Many men who report "feeling better" on magnesium have had an improvement in sleep quality. That is a genuine benefit — it just is not the one on the label, and sleep is where the real hormonal evidence lives anyway.
- Read the funding. A supplement trial paid for by the company selling the supplement is not automatically wrong, but it is a reason to look for a second study. In this case there is one, and it disagrees.
The summary: zinc and magnesium are real nutrients that matter, and the specific claim that supplementing them raises testosterone in men who are not deficient came back empty in the better trial.
